Understanding Cutoffs:
A cutoff is the concentration a drug has to reach before the strip reads positive. These are standard rapid-screen cutoffs. Federal lab cutoffs set by HHS are different for some drugs, which is one more reason a positive cup result goes to a lab for confirmation.

Easy steps to use the 5 panel urine drug test cup
1 - Collect
Have the donor urinate into the cup up to the fill line. Close the lid until it clicks shut. Check the temperature strip within 4 minutes: it should read between 90 and 100°F.
2 - Wait
Set the cup on a flat surface. If your cup has a peel-off label over the result window, remove it now. Wait 5 minutes and don't move the cup.
3 - Read
Read the strips at 5 minutes. Don't interpret results after 10 minutes, because lines can change. Then compare the adulterant pads with the color chart on the label.

Positive
Only the control line (C) appears, with no line at the test region (T). The drug is at or above the cutoff. Treat this as a preliminary positive and send the sample to a lab for GC-MS or LC-MS/MS confirmation before you act on it.
Negative
Two lines appear, at C and T. Any visible T line counts as negative, even a very faint one. The drug is below the cutoff level.
Invalid
No control line appears. The strip didn't run correctly. Throw the cup away and retest with a new one. If it keeps happening, call us with the lot number.

DOT vs Non-DOT 5 Panel Drug Screening
Both kinds of 5 panel drug screening look at the same core drug classes, but the rules behind them are very different.
DOT 5 Panel Drug Test
DOT testing is required for safety-sensitive transportation workers covered by the FMCSA, FAA, FRA, FTA and PHMSA, such as truck and bus drivers, pilots and rail crews. Under 49 CFR Part 40, the urine is collected with a federal custody and control form and tested at an HHS-certified laboratory, and a Medical Review Officer (MRO) reviews the results. The DOT panel covers marijuana, cocaine, amphetamines (including methamphetamine and MDMA), opioids (codeine, morphine, heroin, hydrocodone, hydromorphone, oxycodone, oxymorphone) and PCP. An instant cup cannot be used in place of a DOT lab test.
Non-DOT 5 Panel Drug Test
Non-DOT testing covers everyone else: private employers, staffing agencies, clinics, treatment centers, schools and many court programs. You choose the drugs, the cutoffs and the method. A “forensic use only” test has not gone through FDA 510(k) clearance for clinical use. It is intended for legal and compliance settings, where rules of evidence, chain of custody, and laboratory confirmation help protect the integrity of the result. The 5 panel cups on this page are CLIA-waived and FDA cleared for point-of-care screening.

Forensic results answer one question: did someone follow the rules of their program (probation, sober living, custody)? They don't diagnose a substance use disorder or guide treatment. Most courts require a GC-MS or LC-MS/MS lab confirmation before a screening result is accepted as evidence.

A faint line at T, alongside the control line, is a negative. Line darkness doesn't show how much drug is present. Any visible line means the drug is below the cutoff.

It is a reliable screening test when you read it at the right time, but any positive is preliminary. Send non-negative samples to a lab for GC-MS or LC-MS/MS confirmation before making a hiring, legal or treatment decision. The sensitivity figures for each strip are in the package insert.

Yes. The 90 mL OXY cup tests for AMP, COC, MET, OPI 300 and OXY, with no THC strip. It's popular in states where employers have dropped marijuana testing.
No. Synthetic cannabinoids (K2/spice) and kratom need dedicated strips. A standard 5 panel cup won't pick them up.
It shows the sample's temperature within the first 4 minutes. Fresh urine reads 90–100°F. A reading outside that range suggests the sample may not be fresh or real.

The temperature strip and the adulterant pads can flag many substituted samples, because synthetic urine often falls outside normal human ranges for temperature, creatinine, pH or specific gravity. For high-stakes testing, use observed collection and lab confirmation.
