What Is EDDP?

EDDP (2-ethylidene-1,5-dimethyl-3,3-diphenylpyrrolidine) is the major metabolite formed when the liver breaks down methadone. It is excreted in urine along with methadone itself.

Why Test for EDDP?

Opioid treatment programs and methadone clinics use EDDP testing to verify medication compliance. A methadone-only test can be fooled if methadone is dropped directly into a urine sample, but EDDP only appears after methadone has passed through the body. EDDP testing is also helpful when other medications change how quickly a patient metabolizes methadone.

How to Use the EDDP Dip Card

Remove the dip card from the sealed foil pouch and take off the cap.
Dip the exposed test strip into the urine sample for 10–15 seconds.
Allow the urine to migrate up the test strip.
Replace the cap and lay the dip card flat on a clean, dry, non-absorbent surface.
Read the results at 5 minutes. Do not read after 10 minutes.

How to Read EDDP Dip Card Results

Positive

Only one colored line appears, in the control region (C). No line appears in the test region (T). This means EDDP was detected at or above 100 ng/mL.

Negative

Two colored lines appear: one in the control region (C) and one in the test region (T). Any line in the T region, even a very faint one, counts as negative.

Invalid

No line appears in the control region (C). Discard the result and repeat the test with a new dip card. If the problem persists, stop using the lot and contact 12 Panel Now.

What is EDDP and why test for it instead of methadone?

EDDP is the main metabolite of methadone. A standard methadone (MTD) test detects the drug itself, which can be added to a urine sample to fake compliance. EDDP is produced only when methadone is metabolized by the body, so a positive EDDP result shows the methadone was actually taken.

What does it mean if methadone is positive but EDDP is negative?

This pattern can be a sign that methadone was added directly to the urine sample (spiking) instead of being taken. Because there can be other explanations, review the result together with the patient’s history and confirm it with a laboratory test.

How long does EDDP stay in urine?

EDDP is typically detectable for about 3–4 days after the last methadone dose. Patients on daily maintenance dosing can remain positive for up to about a week. Detection time varies with dose, duration of treatment and individual metabolism.

Will heroin, oxycodone or fentanyl show up on an EDDP test?

No. The EDDP test is designed to detect the methadone metabolite. Heroin, morphine, codeine, oxycodone and fentanyl each require their own test – pair this card with OPI or OXY dip cards, or choose a multi-panel dip card.

How do I read a very faint line?

Any visible line in the test region (T), even a very faint one, means the result is negative. The result is positive only when a line appears in the control region (C) and no line appears in the test region (T).

Is a positive result final?

No. This is a rapid screening test that gives a preliminary result. Confirm non-negative results with a laboratory test such as GC/MS or LC-MS/MS before making treatment, employment or legal decisions.

Do I need a CLIA certificate to use this test?
